Definition of Androgen Receptor
Encoded by human AR Gene (NR3 Family), the androgen receptor transcription factor regulates gene expression, cell proliferation, and differentiation. AR has 3 domains: N-terminal modulating (exon 1), DNA-binding (two C4 Zinc fingers, exons 2-3), and C-terminal hormone-binding (5 remaining exons). Polymorphic N-terminal poly-Gln tracts bind RAN, enhancing transactivation -- decreased with increased poly-Gln length. An 87-kD (AR-A) isoform lacks a normal N-terminus, seen in the 110 kD (AR-B) form -- A and B may differ in function. Ligand bound receptor translocates to the nucleus, homodimerizes, binds DNA, and stimulates transcription. Polyglutamine expansion causes Kennedy's disease. Other mutations are associated with androgen insensibility syndrome. (from LocusLink, Swiss-Prot, OMIM, and NCI)
